采用比较分子场分析法(CoMFA)研究了55个四氢吡啶类毒蕈碱受体激动剂的三维定量构效关系(3D-QSAR), 建立了具有较强预测能力的3D-QSAR模型. 所得模型的交叉验证相关系数(q2)为0.507, 常规相关系数(R2)为0.982 , 标准方差为0.218, 说明系列化合物分子周围立体场和静电场的分布与生物活性间存在良好的相关性. 模型不仅很好地预测了训练集和测试集化合物的活性, 而且为设计活性更高的受体激动剂提供了理论依据. 相似文献

On-line hyphenation of capillary electrophoresis with flame-heated furnace atomic absorption spectrometry for trace mercury speciation 总被引:2,自引:0,他引:2
Capillary electrophoresis (CE) was directly interfaced to flame-heated furnace atomic absorption spectrometry (FHF-AAS) via a laboratory-made thermospray interface for nanoliter trace element speciation. The CE-FHF-AAS interface integrated the superiorities of stable CE separation, complete sample introduction, and continuous vaporization for AAS detection without the need of extra external heat sources and any post-column derivation steps. To demonstrate the usefulness of the developed hybrid technique for speciation analysis, three environmentally significant and toxic forms of methylmercury (MeHg), phenylmercury (PhHg), and inorganic mercury (Hg(II)) were taken as model analytes. Baseline separation of the three mercury species was achieved by CE in a 60 cm long x 75 microm inner diameter fused-silica capillary at 20 kV and using a mixture of 100 mM boric acid and 10% v/v methanol (pH 8.30) as running electrolyte. The precision (relative standard deviation, RSD, n = 7) of migration time, peak area and peak height for the mercury species at 500 microg x L(-1) (as Hg) level were in the range of 0.9-1.2%, 1.5-1.9%, and 1.4-2.0%, respectively. The detection limit (S/N = 3) of three mercury species was 3.0 +/- 0.15 pg (as Hg), corresponding to 50.8 +/- 2.4 microg x L(-1) (as Hg) for 60 nL sample injection, which was almost independent on specific mercury species. The developed hybrid technique was successfully applied to the speciation analysis of mercury in a certified reference material (DORM-2, dogfish muscle). 相似文献

A novel, simple and accurate fingerprint method was developed using high-performance liquid chromatography-photodiode array detection (HPLC-DAD) for the quality control of Hypericum japonicum thunb (Tianjihuang), a Chinese herbal medicine used for the treatment of several bacterial diseases, infectious hepatitis, gastrointestinal disorder, internal hemorrhage and tumors. For the first time, the feasibility and advantages of employing chromatographic fingerprint were investigated and demonstrated for the evaluation of Tianjihuang by systematically comparing chromatograms with a professional analytical software recommended by State Food and Drug Administration (SFDA). Our results revealed that the chromatographic fingerprint combining similarity evaluation could efficiently identify and distinguish raw herbs of Tianjihuang from different sources. The effects resulted from collecting locations, harvesting time and storage time on herbal chromatographic fingerprints were also examined. 相似文献

Geometry optimizations are performed at the DFTB3LYP6-311+G* level. Four intriguing coupling modes, totally eight stable structures are found in the potential energy surfaces of the water-assisted coupling of imidazole dimer radical cation. In these isomers, the water molecules are embedded between two imidazole moieties, and the oxygen atom is tridentate or quadridentate, respectively. The distinct redshifts of the vibrational frequencies of the O-H...N and N-H...O type H bonds indicate the strong interaction of two imidazole rings of respective isomer. Inspection of the highest occupied molecular orbital predicts the alterations of the geometry structures on oxidation and reduction. The low barrier of the fragment rotation demonstrates that the isomerization processes by experiencing the distinct transition states are easy to fulfill, especially for those with O-H...N and C-H...O H bonds. Both the energy difference of the 0 degrees-cis and 180 degrees-trans orientation and the barriers of the fragment rotation are lowered by the water assisting. The range of the zero point vibrational energy correction indicates that the influence on the complexes with N-H...O and O-H...N H bonds (0.13-0.17 kcal/mol) is more significant than those with O-H...N and C-H...O H bonds (+/-0.03 kcal/mol). The dissociation energies of these isomers indicate that the charges transfer easily through water in the dissociation process and then are distributed mainly over the imidazole ring connecting with water molecule. The isomer with proton transfer between imidazole fragments is the most stable one. 相似文献

The reaction of the alkyne HCCC(O)OMe with 7 sep 1,2-(Cp*RuH)2B3H7 leads to hydroboration plus hydroruthenation to produce nido-1,3-mu-Me{C(O)OMe}C-1,2-(Cp*Ru)2B3H7, a compound with an exocluster ruthenium-boron mu-alkylidene that exists in two isomeric forms. Both isomers undergo rearrangement with intramolecular chelation of the carbonyl oxygen at a boron site, thereby opening the cluster and generating arachno-2,3,-mu(C)-5-eta1(O)-Me{C(O)OMe}C-1,2-(Cp*Ru)2B3H7. Further heating leads to deoxygenation of the carbonyl fragment by a boron center concurrent with insertion of the carbon atom into the metallaborane cage to give nido-1,2-(Cp*RuH)2-3-HOB-4-MeC-5-MeOC-BH3. 相似文献

Six novel μ-oxamido binuclear complexes, namely Cu(axpn)Ln(L)2(ClO4)3 (Ln: Eu, Gd, Tb, Nd, Ho, Er), where oxpn is N,N'-bis(3-aminopropyl) oxamido, L denotes 5-nitro,10-phenanthroline (abbreviated as NO2-phen), have been synthesized and characterised. The magnetic susceptibility of complexes Cu(oxpn)Gd(NO2-phen)2(ClO4)3.2H2O was measured over the 4–300 K and the observed data were successfully simulated by equation based on spin Hamiltonian operator (H = -2J1 · S2), giving the exchange integral J(Cu-Gd)=-1.62 cm?1. Crystal growth of calcium oxalate (CaC2O4) in bulk aqueous solution, reverse microemulsion of p-octyl polyethylene glycol phenylether (OP)/iso-octyl alcohol (IOA)/cydohexane/water and above microemulsions containing different kinds of amino acids, such as aspartic acid (Asp), tyrosine (Tyr) and tryptophan (Trp) were studied. The results indicated that different crystallization types of the crystals, which were calcium oxalate monohydrate (COM), calcium oxalate dihydrate (COD) and calcium oxalate trihydrate (COT), existed in bulk aqueous solution. But CaC2O4 growth mainly paralleled with (1 01) plane of COM in reverse microemulsion because of the induction of surfactant at water/oil interface. After adding amino acids into microemulsions, the growth of CaC2O4 crystals mainly influenced by the varieties of amino acids and the pH values of the amino acid aqueous solution. When pH values of the solutions was higher than isoelectric points of amino acids, CaC2O4 crystal paralleled with (1 01) plane of COM more easily with the addition of Trp, Tyr, Asp in turn; however, when pH of the solutions was lower than isoelectric points of Trp, CaC2O4 crystal growth paralleled with (020) face of COM. It is obviously that amino acids, pH values of the solutions and surfactant played important roles in the process of crystal growth of CaC2O4 in the microemulsions.
